Circuit to report transcription in vivo (alternate design)

This part places a green fluorescent aptamer, F30-Broccoli, after a constitutive promoter. This circuit is designed to report general transcriptional levels in vivo in E. coli. As the fluorescence output of this circuit is a direct cause of transcription, it can be used as a measure of a second, test circuit's impact on the host's capacity to transcribe DNA.

Once the correct aptamer dye has been applied to cells transformed with this circuit, a fluorescence output will be produced from the aptamer. As the circuit does not include an RBS, this circuit is able to detect decreases in burden on the transcriptional level. Therefore, based on the amount of aptamer transcribed, the fluorescence output will fluctuate, providing a measure of the test circuit’s impact on the host.

This is an alternative design to BBa_K3773511; BBa_K3773512 instead uses the promoter P70a.
